ABSTRACT

In the last few decades, hydroxyapatite (HA) has become one of the most highly prized biominerals in the biomedical industry for orthopedic and dental applications. The focus of this research was to synthesize biomimetic HA from Tridax procumbens (TP) leaf extract and investigate their antibiofilm properties. The HA was made using the sol-gel method and the HA-TP biocomposite was made by precipitation method. The d.nm size of HA and HA-TP biocomposite was determined as 193.28 and 258.14 d.nm, respectively. The zeta potential of HA and HA-TP biocomposite was determined as ?21.2 and ?18.3 mV, respectively, and found highly stable. The FTIR study revealed that phytochemicals of TP were successfully impregnated into HA-TP biocomposite. The HA and HA-TP biocomposite were found spherical and agglomerated from SEM analysis. In HR-TEM analysis, the average diameter of the HA and HA-TP biocomposite were 16.57 – 64.22 nm and 51.71 – 138.68 nm, respectively. According to the EDX analysis, HA is primarily composed of calcium, oxygen, and phosphate, whereas, HA-TP biocomposite is primarily composed of calcium, phosphate, oxygen, and carbon. In the antioxidant assay, the IC50 value (concentration required to scavenge 50% of free radicals) of HA-TP biocomposite was determined as 156.69 ± 14.02 and 180.21 ± 12.84 µg/mL in DPPH and ABTS free radical scavenging assays, respectively. The MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration) of as-synthesized HA-TP biocomposite against Staphylococcus aureus – ATCC 13565 and Escherichia coli – MTCC 41 were observed as 181.09 ± 21.47 and 317.30 ± 41.03, and 157.59 ± 32.18 and 264.03 ± 21.58 µg/mL, respectively. The as-synthesized HA-TP biocomposite has detrimentally affected the biofilm formation of both the tested bacteria S. aureus – ATCC 13565 and E. coli – MTCC 41. The study concluded that the as-synthesized HA-TP biocomposite could be highly helpful in the biomedical field for alleviating oxidative-stress-related disorders and inhibiting microbial biofilm formation.

ABSTRACT

Objective: The objective of the present study is to evaluate the effect of Phthalate analogues of diclofenac in Freund’s complete adjuvant (FCA) induced Arthritis in the rat. Methods: Twenty four female albino wistar rats were enrolled in this study and are divided into 4 groups (six each). The groups were designed as follows: Group I: vehicle control, Group II: arthritic control, Group III: diclofenac treated, Group IV: phthalate analogue of diclofenac treated. Various assessments such as anti-arthritic activity, biochemical estimations, haematological parameters, ulcerogenesis, radiological and histopathological studies were evaluated. Results: Arthritic control group exhibited significant increase in the level of paw volume, arthritic score (p<0.0001), Serum glutamic pyruvic transaminase (SGPT) (p<0.001), Serum glutamic oxaloacetic transaminase (SGOT) p<0.01), rheumatoid arthritis factor, C-reactive protein (CRP), White Blood Cells (WBC), Creatinine and uric acid and a significant decrease in Red Blood Cells (RBC). Increased swelling of joints, bony destruction and profound ulceration were observed in the Arthritic control group. All these conditions were reversed in diclofenac and phthalate analogue of diclofenac groups. Conclusion: We conclude that phthalate analogue of diclofenac shows potent anti-arthritic activity with milder ulceration when compared to diclofenac treatment.

Background: Vascular anomalies comprise a widely heterogeneous group of lesions. Diagnosis and management of these lesions present challenges to the surgeons, radiologists and histopathologists. Accurate classification of these lesions results in appropriate therapy. Aim of the study is to study the role of histopathology and histochemical stain in the diagnosis of vascular malformationMethods: The present study is a hospital based observational study on vascular malformations over a period of three years from 2016 to 2018 done in department of Pathology and Plastic Surgery at tertiary care centre, Visakhapatnam.Results: Out of 107 specimens of vascular anomalies received, 72 cases were vascular neoplasms, 35 were vascular malformations. Majority of the vascular malformations were seen in the cervicofacial region (43%) followed by extremities (37.1%). Most common malformation in the present study was arterio venous malformations (60%) followed by venous malformations (22.8%). Verhoeff’s VanGieson stain demonstrated discontinuity of internal elastic lamina in cases of arteriovenous malformation and helped in differentiating the lesions from capillary malformation and vascular tumors.Conclusions: Histopathology, special histochemical stains along with imaging features can be used to reduce diagnostic difficulties and in helping proper management of vascular malformations.

ABSTRACT

Background: Rhinosporidiosis is a chronic granulomatous disease caused by aquatic parasite Rhinosporidium seeberi belonging to group of fish parasite Mesomycetozoa. It commonly affects nose and nasopharynx. It is one of the endemic disease is in India. Materials and Methods: This is a prospective study of distribution pattern and management of 12 cases of rhinosporidiosis in our institute(GIMSR)from the region of Visakhapatnam, Vizianagaram and Srikakulam districts of Andhra Pradesh, India. This is also to study the pattern of involvement according to age, sex, site, laterality, and their management. It emphasizes the importance of excision under local anaesthesia once the stalk of the lesion is identified. Results: Our study of 12 patients were shown slightly male preponderance, around the age of 11–20 years, with a clear cut history of having a bath in contaminated pools and rivers . Nasal obstruction & epistaxis are the predominant symptoms. The majority of cases had been excised endoscopically under local anaesthesia with less bleeding and minimal recurrence rate. It also reveals the importance of general anesthesia when the lesions involving posterior aspect of nasal cavity, nasopharynx to prevent aspiration in to lungs and in children who don’t cooperate for local anaesthesia. Conclusion: Endoscopic identification of stalk is mandatory before excising the lesion under local anaesthesia. The bleeding is less when excision done with diathermy cautery.

Background: The Yale observation scale (YOS) is an illness severity helps to diagnose bacteremia based on simple noninvasive clinical signs and symptoms. The aim of the present study was to assess the utility of YOS as a predictor of bacterial infection in febrile children aged 3 to 36 months.Methods: This prospective observational study was conducted on 200 children aged 3 to 36 months presenting with fever, at the Institute of Child Health and Hospital for Children during the period from April 2016 to September 2016. Rectal temperature was taken for all children. Clinical examination was done as required based on the YOS and scores were given accordingly at the time of initial presentation of the child before invasive investigations. All the observation was assessed statistically and receiver operating characteristics (ROC) curve was performed to analyze the sensitivity of the YOS.Results: Highly significant correlation (p=0.0001) was found to exist between the age of the child, duration of the fever, higher body temperature >104, WBC count, ANC and improved condition of patient with higher YOS. ROC curves showed that the sensitivity and specificity of YOS at the best cut off value of 14.5 was found to be 97% and 79.6% respectively.Conclusions: YOS is very good tool for predicting bacteremia in young febrile children based on simple non-invasive clinical signs and symptoms. The findings ruled out by YOS aids in the immediate and early management of bacterial infections before the arrival of the results of the biochemical diagnostic tests.

Background: In spite of distinctive improvements in nutritional therapy, malnutrition and growth retardation remained as challenging significances of chronic liver disease (CLD) in children. The present study was done to evaluate the severity and frequency of malnutrition using anthropometry in CLD children.Methods: The study included 50 children with CLD attending the OPD of Institute of Child Health and Hospital for Children, Egmore, Chennai during the period from April 2016 to September 2016. Physical growth and nutritional status in the patients were evaluated using anthropometric parameters and Z-scores.Results: The incidence of malnutrition in the children using height for age was found to be 90%, weight for age- 84%, BMI for age- 40%, MAC for age- 88%, TST for age- 88%. SGA showed 96% malnutrition. Weight for age and body mass index were influenced by this fluid retention (P=0.002 and P=0.007 respectively). Whereas height for age, MAC and TST were not influenced by fluid retention (P=0.321, P=0.371 and P=0.031 respectively).Conclusions: Assessment of nutritional status in CLD children can be easily diagnosed by anthropometric measurements. Early diagnosis of malnutrition will decrease the related morbidity and mortality in children.

Pod fly, Melanagromyza obtusa is an important emerging pest and a major constraint to increase the production and productivity of pigeonpea. The concealed mode of life of pod fly within the pod makes it difficult to control. Hence, host plant resistance is an important tool for the management of this pest. Therefore, a set of forty genotypes were screened for resistance to pod fly under field conditions and characterized for morphological and biochemical traits in the pods. The correlation studies revealed that, among morphological and biochemical constituents of pigeonpea, pod length (r=0.389*), pod width (r=0.380*), protein content (r=0.857**), total carbohydrates (r=0.782**), reducing sugars (r=0.848**) and total free amino acids (r=0.832**) in the pod walls were positively correlated with per cent pod damage, whereas pod wall thickness (r= -0.762**), trichome density (r= -0.745**) and phenol content (r= -0.871**) had significant negative correlation with pod fly damage. Therefore, these traits can be used as phenotypic and biochemical markers to identify pigeonpea genotypes with resistance to M. obtusa, and use in pod fly resistance breeding program.

A series of blend microspheres were developed from gelatin and hydroxypropyl cellulose (HPC) by emulsion crosslinking method employing glutaraldehyde (GA) as a crosslinker. Valganociclovir hydrochloride (VHCL), an anti HIV drug, was loaded in to these microspheres via insitu method. These microspheres were characterized by Fourier transform infrared spectroscopy (FTIR), to confirm the formation of crosslinking and absence of chemical interactions between drug, polymer and crosslinking agent. Further the microspheres were characterized by scanning electron microscopy to study the surface morphology of the microspheres and observed that the microspheres have smooth surface with spherical structure and no phase separation. The microspheres with the average particle sizes ranging from 614.5μm to 693.4μm were obtained. X-ray diffraction (X-RD) studies were performed to understand the crystalline nature of drug and its uniform distribution into blend microspheres. An in vitro release study was performed in phosphate buffer solution (pH-7.4) at 370C. The release rates were fitted to an emperical equation to understand the diffusion parameters, which indicate non-Fickian or anomalous trend release of VHCL. Further the results indicated that the release of drug was found for more than 12 h.

Five promising early sugarcane clones (2006 T3, 2006 T8, 2006 T19, 2006 T23 and 2006 T36) from yield trials along with two checks (Co C 671 and Co 94008) were evaluated for identification of sugarcane clones suitable for delayed harvest and crushing” at Agricultural Research Station, Perumallapalle, Chittoor, Acharya N.G. Ranga Agricultural University, Andhra Pradesh, during 2011-2012. The data on different growth parameters viz., Germination per cent, Plant height, SCMR values and Leaf area index were recorded at different crop growth periods. The highest germination count was recorded with 2006 T36 followed by Co C 671 and Co 94008, whereas the lowest count was recorded with 2006 T23. Among the clones 2006 T3 and 2006 T36 recorded the highest plant height at different crop growth periods (90, 120, 240 DAP and at harvest) along with two standards (Co C 671 and Co 94008), while the clone 2006 T23 recorded the lowest plant height. The highest SCMR values and LAI was recorded with Co C 671, Co 94008 followed by 2006 T36 and 2006 T3 at different crop growth periods (60, 90, 120 and 150 days), whereas, the clone 2006 T23 recorded the lowest SCMR and LAI values. The data recorded on different yield attributes viz., cane diameter (cm), cane length (cm), number of millable canes and cane yield at harvest showed that 2006 T36 and 2006 T3 recorded the higher cane yields along with checks, while the clone 2006 T23 recorded the lowest cane yield.

ABSTRACT

A reliable method of plant regeneration has been achieved from Axillary buds. Shoots appeared from explants when cultured on Murashige and skoog (MS) medium supplemented with BAP (Benzyl amino purine), Napthalene acetic acid (NAA) and Kinetin at various combinations. Elongated shoots were rooted with 70.6% rooting frequency in MS medium with indole buteric acid (IBA) at 1.0mg/l. The rooted plantlets were established well in soilrite mixture medium with 91% success and days taken for acclimatization were 12.8 days. This protocol was used in Agrobacterium mediated transformation. The transformation was carried out using the Agrobacterium strain LBA4404 containing the binary vector pCAMBIA2301 harboring npt II as selectable marker and GUS as reporter gene.

An efficient direct shoot bud differentiation and multiple shoot induction from shoot tip explants of pigeon pea (Cajanus cajan L.) has been achieved. The frequency of shoot bud regeneration was influenced by the type of explants, genotype and concentrations of cytokinin. Explants viz. shoot tip isolated from 10 day old seedlings showed better explants response Explants were cultured on Murashige and skoog (MS) medium augmented with different concentrations of BAP and NAA. Among the various concentrations tested, 2.0mg/l BAP (Benzyl amino purine) and 0.1 mg/l Napthalene acetic acid (NAA) were found to be the best for maximum shoot bud differentiation. Percentage, as well as the number of shoots per explant showing differentiation of shoot buds was higher on MS media supplement with BAP and optimal BAP concentration for shoot regeneration was 2mg/l. The elongated shoots were successfully rooted on MS medium containing different concentrations of auxins. Among them indole buteric acid (IBA) at 1.0mg/l induced maximum frequency of rooting. Regenerated plants were successfully established in soil where 91% of them have been developed into morphologically normal and fertile plants. This method can thus be advantageously applied in the production of transgenic pigeon pea plants.

The article reports on a development of RP-HPLC method for the quantitative determination of Levetiracetam in tablet dosage forms. The chromatographic separations were performed using Phenomenex C18 (250 mm x 4.6 mm i.d, 5 μm particle size) column at 40 ºC temperatures. The optimum mobile phase consisted of methanol, water and acetonitrile in the ratio of 30:10:60. Auto sampler 20 μl was used and kept at 15 ºC temperature. Analysis was done with flow rate of 1.0 ml/min at 212 nm ( max of Levetiracetam) wavelength by using photodiode array (PDA) detector. The drug was analyzed for acid, alkaline, oxidative, hydrolytic, photolytic and thermal degradation studies. The standard calibration curve was plotted for the drug and results showed that the drug was linear (r2 = 0.999) in the concentration range between 0.01 – 1.5 μg/ml. The results of stress testing undertaken according to the International Conference on Harmonization (ICH) guidelines reveal that the selected method is selective and stability-indicating for determination of levitiracetam in pharmaceutical formualtion.

Production of rice--the world's most important crop for ensuring food security and addressing poverty will be defeated as temperatures increase in rice-growing areas with continued climate change. Climate change needs us to look at various alternatives for more drought tolerant and tougher strains and to develop a technique to screen a large number of genotypes for high temperature tolerance. Adapting temperature induction response (TIR) technique 100 rice genotypes were screened for thermotolerance. Significant variation for acquired thermotolerance was observed in 100 rice lines. From the 100 genotypes 30 were exhibits themotolerance to induced high temperature.

Climate change needs us to look at various alternatives for more drought tolerant and tougher strains. Rice (Oryza sativa L.) is the most important food crop of the world; drought stress is a serious limiting factor to rice production and yield stability in rainfed areas. In order to design efficient varieties with virtues of drought tolerance and high yielding ability is necessary. Root system plays an important role under drought conditions. Among all the genotypes NLR 33671 showed highest root length (27.6cm) under moisture stress condition followed by NLR 3010 (24.5cm), NLR 40059 (23.6 cm), TELLAHAMSA (23.5 cm) ,NLR 40049 (23.5), NLR 3098 (23.5) were showed significantly superior mean values than remaining genotypes for most of the root traits included in this study. Therefore, these genotypes can be considered as drought resistance varieties. Whereas NLR 40054 (14.35 cm), BPT 5204 (14.55 cm), NLR 40045 (15 cm) and NLR 30491 (15.19cm) recorded lowest root length these can be consider as susceptible ones. The effects of drought stress on rice productivity were explicitly parameterized and addressed in the present study.
